Neuron subtype-specific molecular profiling and gene editing in a mouse model of cocaine addiction

چکیده

Addiction is a complex and devastating disease that hijacks homeostatic reward signaling, resulting in progressive loss of control over drug consumption and aversive withdrawal symptoms following prolonged abstinence. Recent studies have suggested that epigenetic modifications, particularly DNA methylation and histone methylation, regulate the long-term storage and maintenance of memory, and that defects of certain epigenetic factors affect drug-induced behaviors. We therefore hypothesize that epigenetic modifications in midbrain dopamine (DA) neurons play essential roles in regulating behavioral responses to drugs of abuse. However, cell heterogeneity in brain still represents a technical hurdle that impedes molecular profiling and editing at neuronsubtype resolution.

Efficient Production of Biallelic RAG1 Knockout Mouse Embryonic Stem Cell Using CRISPR/Cas9

Background: Recombination Activating Genes (RAG) mutated embryonic stem cells are (ES) cells which are unable to perform V (D) J recombination. These cells can be used for generation of immunodeficient mouse. Creating biallelic mutations by CRISPR/Cas9 genome editing has emerged as a powerful technique to generate site-specific mutations in different sequences. Ob...

Genome-wide Analysis of Chromatin Regulation by Cocaine Reveals a Role for Sirtuins

Changes in gene expression contribute to the long-lasting regulation of the brain's reward circuitry seen in drug addiction; however, the specific genes regulated and the transcriptional mechanisms underlying such regulation remain poorly understood. Here, we used chromatin immunoprecipitation coupled with promoter microarray analysis to characterize genome-wide chromatin changes in the mouse n...

An Unusual Suspect in Cocaine Addiction

Development of drug addiction is extremely complex, but its initiation can be as simple as the flip-flop of glutamatergic receptor subtypes triggered by an "unusual" type of NMDA receptors, as suggested by Yuan et al. (2013) in this issue of Neuron.

Exploring Gene Signatures in Different Molecular Subtypes of Gastric Cancer (MSS/ TP53+, MSS/TP53-): A Network-based and Machine Learning Approach

Gastric cancer (GC) is one of the leading causes of cancer mortality, worldwide. Molecular understanding of GC’s different subtypes is still dismal and it is necessary to develop new subtype-specific diagnostic and therapeutic approaches. Therefore developing comprehensive research in this area is demanding to have a deeper insight into molecular processes, underlying these subtypes. In this st...

Global gene expression analysis using microarray to study differential vulnerability to neurodegeneration

Neurodegenerative disorders such as Parkinson’s disease, motor neuron disease and Alzheimer’s disease is characterized by loss of specific cells within certain regions of the brain. One of the most compelling questions is to determine why specific cell populations are vulnerable to neurodegeneration. We addressed this question by studying global gene expression changes using an animal model of ...
